The Kansas City Chiefs selected Mitch in the second round, 49th overall, in the 2015 NFL Draft. He was drafted for the Pro Football Writers of America All-Rookie Team. Mitch made his official NFL debut playing against the Houston Texans during Week 1 of the 2015 regular season. At the end of that season, he was named to the PFWA All-Rookie Team. In 2017, he missed Weeks 3–7 with a foot injury before returning to the following week’s lineup.

On December 15, 2017, he was placed on injured reserve after re-injuring the foot in Week 14. On March 13, 2019, Mitch signed a four-year $44.5 million contract with the Buffalo Bills, becoming the highest-paid center in the league at the time. He signed a two-year, $19.5 million contract extension with the Bills on March 14, 2022.
