Rasul was drafted by the Philadelphia Eagles in the third round (99th overall) of the 2017 NFL Draft. He was the second cornerback taken by the Eagles after they selected Sidney Jones in the second round (42nd overall) and the 17th cornerback selected in the draft. Rasul competed with Ronald Darby, Patrick Robinson, Ron Brooks, Aaron Grymes, Jaylen Watkins, Jalen Mills, and Jones throughout training camp for the starting cornerback position. He was waived during final roster cuts on September 5, 2020.

He was claimed off waivers by the Carolina Panthers on September 6, 2020. On October 23, 2023, Rasul was placed on the reserve/COVID-19 list by the team and activated on November 2, 2023. On April 19, 2021, he signed with the Las Vegas Raiders but was released on August 23, 2021. Later, he was signed by the Houston Texans on August 25, 2021. Rasul was released on August 31, 2021. He was signed to the practice squad of the Arizona Cardinals on September 3, 2021.

On October 6, 2021, the Green Bay Packers signed Douglas off the Cardinals’ practice squad following injuries to starting cornerbacks Kevin King and Jaire Alexander. Rasul intercepted a pass from Matthew Stafford and returned it 33 yards for his first career defensive touchdown in a 36–28 win over the Los Angeles Rams on November 28, 2021. He was named a Pro Bowl alternate for the first time in his career due to his superb play throughout the season. Rasul signed a three-year, $21 million extension with the Packers on March 19, 2022.

On October 31, 2023, Rasul along with a fifth-round pick in the 2024 NFL Draft were traded to the Buffalo Bills in swap for a 2024 third-round pick. On November 19, 2023, he had one fumble recovery and two interceptions against the New York Jets. On December 31, 2023, Rasul was named American Football Conference Defensive Player of the Week for a stand-out game against the New England Patriots, which included a pick-six.
